I'm about to put a new radio and rear speaker in my 2+2 and wondered if there was any tricks or hints on replacing the rear speaker?

none that I'm aware of, it's not like a modern computerized car. I would recommend disconnecting the battery when changing the radio.

I put the original AM radio & rear speaker into a '68 Catalina I had. The car had the "Sepraphonic" option, which was a dash mounted front/rear fader control. I took out the rear seat & side panels to chase down the original speaker wire. There's just a hot (+) wire, the speakers ground (-) to the vehicle body through the mounting. Just make sure you have good electrical mounting contact (no plastic washers/shims).
