Our 05 non SV6 apparently has a battery drain somewhere. We did the amp test and found that the circuit that has the radio and sliding door had some amp draw when off. I took out the fuse in hopes that the battery wont be draining, well after being parked for one day, the van won't start again. What could be draining it?

You have to go back to the test and with the fuse out, see if you still have a drain. How do you know the battery is still good and the alternator is charging, where they tested?

after you make sure the battery is good check your negative and positive cables,if they have any resistance they will pull down the battery,if they are good next would be the onstar module and the bcm= body control module. you should not have more than a .15 amp draw....and thats after your car is off for 30 min,it takes that long for the modules to go into sleep mode.
